Used as an acid-base indicator
Bromophenol blue – long name 3′,3″,5′,5″-tetrabromophenolsulfonphthalein – can be used as a pH indicator, a dye and a colour marker. It’s usually prepared by gently mixing bromine to phenolsulfonphthalein in glacial acetic acid.
- pH indicator: It changes colour from yellow at pH 3 to purple at pH 4.6, in a reversible reaction. In chemical terms, the reaction is akin to what happens to phenolphthalein, another popular pH indicator.
- Colour marker: Popularly used to monitor progress during electrophoresis (both agarose gel and polyacrylamide gel). As this compound has a slight negative charge, it will travel in the gel closely to the DNA or protein being tested. For example, using a 1% agarose gel in TAE or TBE buffer, bromophenol blue travels at the same speed as a 500 base pair DNA fragment.
- Dye: At pH 7 and above, this compound absorbs red light and transmits blue light, which means solutions are blue. In acidic solutions (pH<7), this dye absorbs ultraviolet light and transmits in the yellow range. Interestingly, at pH 3.6 (when colour changes), this compound has a green or red colour, depending on the concentration. This is called dichromatic colour.
